Rybelsus contains semaglutide, a GLP-1 receptor agonist (glucagon-like peptide-1 receptor agonist).

Mechanism of action

Semaglutide mimics the natural hormone GLP-1, which helps regulate blood glucose levels. It works by:

  1. Increasing glucose-dependent insulin secretion

    • When blood sugar is elevated, it stimulates the pancreas to release more insulin.

  2. Reducing glucagon secretion

    • It decreases the release of glucagon, a hormone that signals the liver to produce glucose.

  3. Slowing gastric emptying

    • Food leaves the stomach more slowly, which reduces the rise in blood glucose after meals.

  4. Reducing appetite and increasing satiety

    • It acts on appetite centers in the brain, helping people feel fuller and often leading to weight loss.

Clinical effect

The combined result is:

  • Lower fasting and post-meal blood glucose levels

  • Improved HbA1c

  • Reduced appetite and body weight in many patients

Rybelsus is the oral tablet formulation of semaglutide and is primarily used for the treatment of Type 2 Diabetes. It is taken on an empty stomach to maximize absorption.
